LIQUID CHROMATOGRAPHY AND MASS SPECTROMETRY METHODS IN MARINE SCIENCES

Course Description 

Chromatography and mass spectrometry are very powerful techniques. Their use in Marine Sciences is currently under expansion. The Analytical and Structural Chemistry Platform of EMBRC.PT has developed several protocols for the identification and quantification of bioactive molecules in marine matrices. These Knowledge Exchange and Training activities will give an overview of liquid chromatography and mass spectrometry methods to researchers and demonstrate their potential in Marine Sciences by running specific examples. Early-career researchers and prospective users are particularly welcomed. Participants are encouraged to send selected samples for analysis during the course. 

Main topics

1. High Resolution Mass spectrometry and Tandem Mass Spectrometry (ESI-IRMSn). Identification of bioactive molecules.
2. Liquid Chromatography with Diode Array Detection (LC-DAD). Identification and quantification of pigments/polyphenols.
3. Liquid Chromatography coupled to Mass Spectrometry (LC-MS). Quantification of sterols using isotope dilution method.

Format 

The course involves a general presentation and discussion of each technique, followed by the run of representative samples, discussion of analytical conditions and interpretation of gathered data. 

Three demonstrations/applications will be shown at CCMAR Laboratories. Each demonstration/application involves a laboratory visit within situ presentation and description of the main components of the equipment, followed by real-time analysis of samples and discussion of methods/results in the classroom, using remote control software to access the instrument’s computer. 

Pre-requisites 

Basic formation in sample preparation procedures and HPLC analysis is recommended. Note: we also recommend participants to bring their own computer for data analysis and processing.

Monday, 28th - High Resolution Mass spectrometry and Tandem Mass Spectrometry (ESI-HRMSn)

9:00 - Welcome Reception

A welcome reception at room 2.88 will initiate the course. An overview of CCMAR main research areas and infrastructures will be given. Attendees will briefly (5 min) present their scientific background and main research interests.

10:30 - Coffee-break

10:45 – Presentation ESI-HRMSn technique. Mass spectrometry and the main components of a mass spectrometer. Applicability of mass spectrometry in the various research areas of Marine Sciences. Fundamentals of high resolution mass spectrometry and tandem mass spectrometry. Identification of bioactive compounds.

12:30 - Lunch

14:00 - Application/demonstration. Visit of ESI-HRMSn laboratory. Analysis of male tilapia urine and supramolecular structures by ESI-HRMSn.

16:30 - Coffee-break

17:00 - Application/demonstration. Analysis of other bioactive molecules by ESI-HRMSn.

 

Tuesday 29th - Liquid Chromatography with Diode Array Detection (LC-DAD)

9:30 - Presentation of LC-DAD technique. Fundamentals of LC-DAD. Main components of an LC-DAD system. Applicability of LC in Marine Sciences.

11:30 - Qualitative and quantitative analysis in LC-DAD.

12:30 - Lunch

14:00 - Application/demonstration. Visit of LC-DAD system. Analysis of pigments/polyphenols by LC-DAD.

16:30 - Application/demonstration. Analysis of polyphenols by LC-DAD.

 

Wednesday 30th - Liquid Chromatography coupled to Mass Spectrometry (LC-MS)

9:30 - Presentation of the LC-MS technique. Why coupling a mass spectrometer to an LC system. Applicability of LC-MS in the various areas of Marine Sciences. The relation between molecular structure and the signals observed under LC-MS.

11:00 - Coffee-break

11:30 - Information from LC-MS traces under different operation modes and conditions. Principles of quantitative analysis by LC-MS.

12:30 - Lunch

14:00 - Application/demonstration. Visit the LC-MS system. Multi Reaction Monitoring (MRM). Development of quantification methods using isotope labeled internal standards.

16:00 - Coffee-break

16:30 - Application/demonstration. Quantification of sterols in plasma by UHPLC-HRMS under MRM.
